As you quite rightly pointed out here, you don't have enough information about the precision fill (e.g. gravimetric fill) of the primary std. For this reaon we have always weighed out from the primary vial to prepare it. The scenario's you have providied here relate to your own secondary std, which needs to be assayed by taking 4 individiual weighings and hence preparations of it. The primary standard should be weighed in duplicate i.e. duplicate preparations as a minimum unless you are directed otherwise in the monograph / in-house method.
