Strips are usually placed in the chamber or affixed or an object to be sterilized. The use of strips is not to validate the sterility of a medium but to validate the sterilizing efficiency of the process itself.

If you want to validate the sterilization of a liquid you may both use strips in the chamber and to use a "contaminated" liquid representing the worse case that can happen later on. The "holding time" is the time this liquid is awaiting before to be processed : so, during this time the different microorganisms that are present into the liquid will grow. For a longer holding time you will have a higher bioburden in the liquid. So this will represent your worst case. After the sterilization run you will test the sterility of the liquid on one hand and, on the other hand, you will very that there is no surviving spore on the strips (strips or spores suspension is not a matter : you can use the most appropriate for your purpose).
